IQA Center Laboratory SIV Viral Load Testing

Shared Resource Summary:

The IQA Center Laboratory SIV Viral Load Testing Shared Resource offers testing of non human primate samples for SIV viral load, using the ASI StepOnePlus Real Time PCR System. The facility has established CLIA and CAP accreditation and works under GLP for viral marker core testing. This Shared Resource also provides HIV serology (ELISA and Western blotting), HIV RNA viral load (Roche COSAS) and subsequent HIV acute infection disease staging (e.g., Fiebig).
 
For SIV, the lab has established a de novo SIV, SHIV and SMM viral load assay using the Applied Siosystems StepOnePlus platform. This assay detects SIVmac239, SIVmac251, SHIVsf-162p3, SHIV89.6/89.6p, SIVmac1A11 and SMMPSJ viral strains. Viral RNA extraction is done using the Qiagen QIAamp Viral Mini Kit. RNA is reverse transcribed to cDNA using the Invitrogen SuperScript III. Real-time PCR is set up with the ASI Gene Expression Mastermix and primers specific to the SIV-gag gene. A fluorescently labeled Taqman probe is used to monitor the reaction.

Services Provided: 

SIV viral loads on non-human primates samples.

  • SIV viral load assay for non-human primate samples on a per sample basis.
  • For plasma samples, at least 300mL of sample is needed. 
For other body fluids, please call before sending samples.

 

Protocols and Documents:

  • An IQAC Laboratory procurement sheet must be provided with samples submitted for testing.
